What Determines a Molecule's Polarity?

Polarity arises from differences in electronegativity between bonded atoms. When atoms with significantly different electronegativities form covalent bonds—such as in water (H₂O) or ammonia (NH₃)—electrons are unequally shared, creating dipole moments. The overall molecular polarity depends on both bond polarity and molecular geometry. For example, carbon dioxide (CO₂) has polar bonds but is nonpolar overall due to its linear symmetry, whereas water is bent and thus polar.

How Does Polarity Affect Solubility and Interactions?

The principle 'like dissolves like' governs solubility: polar substances dissolve in polar solvents (e.g., ethanol in water), while nonpolar substances dissolve in nonpolar solvents (e.g., oil in hexane). In biological systems, polarity influences membrane permeability, protein folding, and ligand-receptor binding. Polar functional groups such as hydroxyl (-OH), amine (-NH₂), and carboxyl (-COOH) enhance water solubility and participate in hydrogen bonding, crucial for biomolecular stability and function.

In chromatography (e.g., HPLC, GC-MS), polarity determines retention times and separation efficiency. Polar compounds elute earlier in nonpolar stationary phases. In drug development, polarity affects bioavailability, metabolic stability, and blood-brain barrier penetration. Regulatory guidelines (e.g., ICH Q6A, USP <1151>) often require polarity assessment via logP (octanol-water partition coefficient) to predict pharmacokinetic behaviour.
