Guanidine hydrochloride acts by competing with intramolecular hydrogen bonds and disrupting the hydrophobic core of proteins. At high concentrations (typically 4–8 M), it effectively unfolds proteins into random coils, preventing aggregation and enabling the study of folding pathways. Its effectiveness is often compared to that of urea, though GdnHCl is generally more potent. The denaturation process is reversible upon dilution or removal of GdnHCl, allowing for refolding of functional proteins.
