Guanidine hydrochloride (GdnHCl) is a salt formed from guanidine and hydrochloric acid. It is a strong denaturant that disrupts hydrogen bonding and hydrophobic interactions within proteins, leading to the unfolding of their native structure. This property makes it invaluable in biochemical research, particularly in the study of protein folding and stability. GdnHCl is widely used in the purification and refolding of recombinant proteins, as well as in the solubilisation of inclusion bodies. It is also employed in the preparation of protein samples for analysis by techniques such as SDS-PAGE and mass spectrometry.

How does guanidine hydrochloride function in protein denaturation?

Guanidine hydrochloride acts by competing with intramolecular hydrogen bonds and disrupting the hydrophobic core of proteins. At high concentrations (typically 4–8 M), it effectively unfolds proteins into random coils, preventing aggregation and enabling the study of folding pathways. Its effectiveness is often compared to that of urea, though GdnHCl is generally more potent. The denaturation process is reversible upon dilution or removal of GdnHCl, allowing for refolding of functional proteins. This reversibility is crucial in biopharmaceutical production and protein engineering workflows.
