An amide is a functional group in organic chemistry characterised by a carbonyl group bonded to a nitrogen atom. Amides are key structural motifs in peptides, proteins, and many pharmaceuticals, offering stability and hydrogen-bonding capability.

What is an amide?

Amides are organic compounds derived from carboxylic acids by replacing the hydroxyl group with an amino group. The general structure is R–CO–NH₂, where R can be an alkyl or aryl group. Amides are known for their stability, particularly under physiological conditions, making them essential in biological systems and synthetic chemistry.

How are amides formed?

Amides are typically synthesised through condensation reactions between carboxylic acids and amines, often with the aid of coupling agents such as EDC or HBTU. In biological systems, amide bonds are formed during protein synthesis via ribosomal translation. The amide bond is planar and exhibits partial double-bond character due to resonance, which restricts rotation and contributes to the rigidity of peptide backbones.

Why are amides important in pharmaceuticals and biotechnology?

Amide linkages are prevalent in drug molecules due to their metabolic stability and ability to form hydrogen bonds, which enhance binding affinity to biological targets. Many active pharmaceutical ingredients (APIs) contain amide bonds, including antibiotics like penicillin and antihypertensives such as lisinopril. In biotechnology, amide chemistry is used in conjugation strategies for antibody-drug conjugates (ADCs) and protein immobilisation on solid supports.
